Bug reproduced on:
- macOS Sierra 10.12.26 -- Safari 10.1.2
- macOS High Sierra 10.13 -- Safari 11.0

Could you please provide a complete test case? This works for me.
Specifically, I get "true" when I open https://www.apple.com/apple-events/september-2017/, and execute this code in Web Inspector console:
document.getElementsByTagName("video")[0].webkitSupportsPresentationMode('picture-in-picture')
